Palestra apresenta tecnologia para o desenvolvimento de testes diagnósticos específicos
A palestra “Microarrays de peptídeos de alta resolução: da pesquisa básica ao desenvolvimento de vacinas, testes diagnósticos e anticorpos monoclonais terapêuticos”, apresentada na última sexta-feira 26/5, pelo representante da Empresa BioAlbra Biotecnologia, Dr. Pedro Simonini, no Centro de Estudos do Instituto Leônidas e Maria Deane (ILMD/Fiocruz Amazônia), trouxe ao público a apresentação de uma plataforma voltada para o desenvolvimento de testes diagnósticos específicos, capaz de diferenciar especificidades dos vírus da Zika, Dengue, Chikungunya e Febre Amarela.
“Em princípio é uma tecnologia que já existe, mas o produto que nós desenvolvemos tem um diferencial: Temos a informação completa do genoma e proteoma do vírus da Zika ou Dengue, Chikungunya, Febre Amarela e conseguimos analisar toda essa informação de uma vez”, explicou Simonini.
A grande flexibilidade dessa plataforma permite sintetizar microarranjos personalizados cobrindo desde pequenas sequências peptídicas até proteínas inteiras ou genomas virais completos. Os microarranjos demandam amostras pequenas de soro e podem ser usados em diversas aplicações, como mapeamento de epítopo e análise de substituição, desenvolvimento de vacinas, monitoramento imunológico, descoberta de biomarcadores, perfil de anticorpos do soro, antes e depois de infecções, administração de vacinas, ingestão de medicamentos, perfil de anticorpos autoimunes no soro de pacientes e perfil de anticorpos relacionados com processos alérgicos.
De acordo com o palestrante, o desenvolvimento da tecnologia permitiu a elaboração de projetos complexos de proteômica high-throughput. Através de exemplos concretos, a palestra teve o objetivo de demonstrar o potencial único e as inúmeras aplicações dessa tecnologia tanto em pesquisa básica como aplicada.
A plataforma tem capacidade de sintetizar desde pequenas sequências peptídicas até proteomas completos, como por exemplo: Dengue, Zika, Chikungunya e Febre Amarela. “A flexibilidade de síntese de peptídeos nos permite adaptar a plataforma para qualquer organismo cujo genoma tenha sido sequenciado. Usando microarranjos de peptídeos de alta resolução podemos, por exemplo, caracterizar e diferenciar a resposta humoral entre virus semelhantes, distinguindo inclusive entre infecção e vacinação”, esclareceu Simonini.
Os Microarrays de peptídeos é um equipamento indispensável para diversas aplicações moleculares. O problema principal dessa tecnologia é a sua produção, pois cada peptídeo necessita ser sintetizado individualmente. Após sua síntese cada peptídeo precisa passar por um processo para se ligar ao suporte do microarranjos.
O processo tem limitações intrínsecas, pois além do custo elevado, não permite a produção de microarranjos de peptídeos de alta densidade. Para superar essas limitações, pesquisadores do German Cancer Reserach Center, em Heidelberg, desenvolveram a tecnologia. Usando o princípio de impressão a laser, os peptídeos são sintetizados diretamente sobre a superfície do microarranjo. Essa tecnologia garante uma flexibilidade única e quase ilimitada para sintetizar microarranjos de peptídeos de alta densidade.
